Bicarbonate-buffered medium for oocyte handling, insemination and overnight fertilisation culture in the Cook sequential system.
What it is and how it's used
Equivalent role to other fertilisation media: rinse, hold, inseminate, culture to the pronuclear check. Equilibrate under oil in CO₂ before use.
Typical failure modes
- Fertilisation drop after lot change or storage excursion.
- pH drift with time outside the incubator.
- Osmolality change in small drops.
What to check first
- Lot/open date vs KPI change; fridge log.
- Equilibration time and time-out during procedures.
- Independent CO₂ check of the incubator.
