G-IVF PLUS (fertilisation medium)
Bicarbonate-buffered medium for oocyte handling, insemination and fertilisation (day 0–1) in the Vitrolife sequential system.
What it is and how it's used
Used for rinsing and holding COCs after retrieval, for conventional insemination, and for overnight culture to the fertilisation check. Bicarbonate-buffered, so it must be equilibrated in the incubator (CO₂) before use and kept under oil when out.
Typical failure modes
- Fertilisation and early cleavage fall after a new lot or after a storage/transit temperature excursion.
- pH drift when dishes are prepared too late (under-equilibrated) or left out of the incubator (alkaline drift within minutes).
- Osmolality creep in small drops in low-humidity incubators over the overnight step.
What to check first
- Lot number and opening date against the date the KPI changed; storage temperature log for the fridge.
- Equilibration time actually achieved before use (dish preparation time vs SOP).
- Time dishes spend outside CO₂ during search, insemination and the fertilisation check.
- Independent CO₂ measurement of the incubator it is equilibrated in.
IFU and practice notes
Follow the manufacturer's IFU for storage, shelf life after opening, equilibration and oil overlay. Change one variable at a time when introducing a new lot.
