What it is and how it's used

One medium, no changeover: dishes are prepared once and embryos left undisturbed for 5–6 days. Reduces handling variables and makes incubator stability and drop osmolality the dominant factors.

Typical failure modes

  • Osmolality rise over 5–6 days in dry incubators or small drops → slowed development, poorer blastocysts.
  • Lot-related changes in blastocyst quality without changes in fertilisation.
  • Prolonged pre-equilibration leading to evaporation before embryos are even loaded.

What to check first

  1. Osmolality of medium after the full culture period in the actual incubator (spot check).
  2. Humidification status of the incubator; drop volume; oil volume and quality.
  3. Lot number versus the KPI change; storage conditions.

Follow the IFU for drop volume, overlay, equilibration and culture duration; verify humidity requirements for your incubator model.
